Epitalon

Epitalon (Epithalon / Epithalone)

Also known as: Epithalon, Epithalone, AGAG, Ala-Glu-Asp-Gly

Overview

A synthetic tetrapeptide based on the natural peptide epithalamin produced by the pineal gland. While primarily known as an anti-aging peptide through telomerase activation, Epitalon plays a significant role in sleep regulation by normalizing melatonin production from the pineal gland, which declines with age. This makes it particularly valuable for age-related sleep disturbances and circadian rhythm dysregulation.

Half-Life
~2-3 hours
Typical Dose
5–10 mg
Frequency
Once daily for 10-20 day cycles
Cycle Length
10-20 days on, 4-6 months off

Administration

SubcutaneousIntramuscularIntranasal

Benefits

  • Restores age-depleted melatonin production for natural sleep regulation
  • Normalizes circadian rhythm in elderly individuals
  • Activates telomerase to lengthen telomeres (anti-aging crossover)
  • Improves sleep onset and sleep quality through pineal gland support
  • Supports immune function and neuroendocrine balance

Side Effects

  • Mild injection site irritation
  • Drowsiness (related to melatonin upregulation)
  • Headache (uncommon)

Mechanism of Action

  • Stimulates pinealocytes to restore melatonin synthesis and secretion
  • Reactivates genes involved in circadian clock regulation
  • Activates telomerase enzyme (hTERT) for anti-aging effects
  • Enhances antioxidant defense enzymes (SOD, catalase) to reduce oxidative sleep disruption

Contraindications

  • Active cancer (telomerase activation could theoretically promote tumor growth)
  • Autoimmune conditions without medical supervision
  • Pregnancy or breastfeeding

Storage & Reconstitution

Store at 2–8°C (36–46°F) for up to 3 months. For long-term storage, keep at -20°C (-4°F). Once reconstituted, refrigerate and use within 21 days.

Research Summary

Developed by Professor Vladimir Khavinson at the St. Petersburg Institute of Bioregulation and Gerontology. A key study in elderly patients (ages 60-80) demonstrated that Epitalon restored evening melatonin peaks to levels resembling those of younger adults, with corresponding improvements in sleep quality and daytime alertness. The 15-year longitudinal study showed improved immune function, cardiovascular health, and endocrine balance alongside the sleep benefits. Its dual role in melatonin restoration and telomere elongation makes it uniquely positioned among sleep-supportive peptides.
