Kisspeptin-10

Kisspeptin-10 (Metastin 45-54)

Also known as: KP-10, Metastin 45-54, Kisspeptin

Overview

A 10-amino acid C-terminal fragment of the kisspeptin family that is a potent agonist of the GPR54 (KISS1R) receptor. Kisspeptin is a master regulator of the hypothalamic-pituitary-gonadal (HPG) axis and plays a critical role in puberty onset, reproductive hormone secretion, and sexual behavior.

Half-Life
~4 minutes (IV bolus); ~28 minutes (SC)
Typical Dose
1–10 mcg
Frequency
single dose or pulsatile infusion in research protocols
Cycle Length
Research protocols vary; typically acute or short-term dosing

Administration

SubcutaneousIntravenous

Benefits

  • Potent stimulator of endogenous LH and FSH release
  • Restores pulsatile GnRH secretion in hypothalamic amenorrhea
  • Enhances sexual arousal and brain limbic activity in neuroimaging studies
  • May improve reproductive function in both sexes
  • Acts as a physiological regulator rather than pharmacological override

Side Effects

  • Mild facial flushing
  • Transient lower abdominal warmth
  • Injection site discomfort
  • Potential ovarian hyperstimulation in women (in clinical settings)

Mechanism of Action

  • Binds KISS1R (GPR54) on GnRH neurons in the hypothalamic arcuate nucleus, triggering GnRH pulse secretion
  • Downstream GnRH release stimulates anterior pituitary gonadotrophs to secrete LH and FSH
  • Activates limbic brain regions (amygdala, cingulate cortex) associated with sexual arousal and emotional processing

Contraindications

  • Hormone-sensitive malignancies (breast, prostate)
  • Pregnancy or breastfeeding
  • Pituitary tumors or disorders

Storage & Reconstitution

Store at 2–8°C (36–46°F) for up to 3 months. For long-term storage, keep at -20°C (-4°F). Once reconstituted, refrigerate and use within 14 days. Avoid repeated freeze-thaw cycles.

Common peptide amounts:1 mg5 mg

Research Summary

Kisspeptin-10 has been studied in multiple Phase I/II clinical trials at Imperial College London and other institutions. Research demonstrates it potently stimulates LH secretion in healthy volunteers and can restore reproductive hormone pulsatility in women with hypothalamic amenorrhea. Functional MRI studies have shown kisspeptin enhances brain processing of sexual and romantic stimuli, suggesting a role beyond pure endocrine regulation. It is being investigated as a potential therapeutic for infertility and low sexual desire.
